چکیده

Objective:The present study has been designed to investigate efficacy Of OG-1 cardioactive principle from chloroform extract of Ocimum gratissimum (Linn.) Leaves on ischaemia and reperfusion-Induced myocardial injury. Methods: Various extracts of O. gratissimum (Linn.) leaves were prepared viz. Pet.ether, chloroform, acetone and methanol. Among all the extracts chloroform extract reduce myocardial injury. The chloroform extract further purified using column chromatographic technique that results four major fractions viz F1, F2, F3 and F4. Among these fractions F4 was found to be hot fraction. OG1 (active principle) was isolated from the F4 using column chromatography technique and evaluated for cardioprotective effect. Results: The present study demonstrated that Chloroform extract of O.gratissimum (Linn.) leaves and further fraction F4 of chloroform extract significantly prevented myocardial infarct size as compared with that of standard ramipril. OG1 (active principle) was isolated from fraction F4 of chloroform extract showing cardioprotective effect at dose level of 50mg/kg body weight as compared with that of standard ramipril (1mg/kg body weight). Conclusion:These results suggest that OG1 is active cardioprotective principle of O. gratissimum (Linn.) chloroform leaves extract.

Exploring the role of dimethylarginine dimethylaminohydrolase-mediated reduction in tissue asymmetrical dimethylarginine levels in cardio-protective mechanism of ischaemic postconditioning in rats

Objective(s): Reperfusion of ischaemic myocardium results in reduced nitric oxide (NO) biosynthesis by endothelial nitric oxide synthase (eNOS) leading to endothelial dysfunction and subsequent tissue damage. Impaired NO biosynthesis may be partly due to increased levels of asymmetrical dimethylarginine (ADMA), an endogenous inhibitor of eNOS. As dimethylarginine dimet...

Effects of hydroalcoholic extract from aerial parts of the sterile stems of Stachys inflata on myocardial infarct size in rats

Recently a potent anti-inflammatory effect of hydroalcoholic extract of the aerial parts of the sterile stems of Stachys inflata has been reported. This study examined whether hydroalcoholic extract isolated from aerial parts of non-flowering stems of Stachys inflata (standardized to contain 4.5% caffeic acid derivatives) reduce myocardial infarct size arising from coronary artery occlusion (30...

Calpain Inhibitors and Modulation of Ischaemia Reperfusion Induced Apoptosis and Necrotic Myocardial Cell Injury

Apoptosis is a crucial event that can initiate ischaemia-reperfusion induced inflammation and subsequent tissue injury. Myocardial ischaemia reperfusion is associated with activation of intracellular death proteases known as calpains. Myocardial ischaemia was induced in isolated rat heart which was subjected to 30 min ischaemia followed by reperfusion for 120min. The effect of calpain inhibitor...

Role of SUMOylation and deSUMOylation of mitochondrial fission proteins in myocardial ischaemia-reperfusion injury

Background Restoration of blood supply to the heart after a prolonged ischaemic episode causes myocardial ischaemia-reperfusion (I/R) injury. Paradoxically, short periods of ischaemia and reperfusion, known as preconditioning, has a potent cardioprotective effect. One key aspect of I/R injury is cytotoxic mitochondrial fission that allows cytochrome c release and promotes apoptotic cell death. ...
